A nucleotide has three components i.e. A nitrogen base, A pentose sugar, A phosphate group. Nitrogenous base is linked to the pentose sugar through N-glycosidic linkage to form nucleoside but when phosphate group is linked to this nucleoside through phosphodiester linkage a nucleotide is formed and when many nucleotides are linked, then a polynucleotide chain is formed. Base pairing confers a very unique property to the polynucleotide chain.

Phage dilution was prepared in trypton broth and two drops of E. coli. were added when bacteriophage affect the bacteria, it causes lytic cycle and form plaque.
